Disease_relevanceMutations in human PARK2/Parkin are associated with the juvenile form of Parkinson disease, adenocarcinomas of the ovary and lung, and susceptibility to Leprosy; the Parkin protein is a RING domain-containing E3 ubiquitin ligase, involved in proteasome-dependent degradation of proteins and protection against cellular and mitochondrial stress; in elegans, cytosolic and ER stress exerts specific toxicity in pdr-1/parkin mutants; the truncated PDR-1(delta aa24-247) allele, when expressed in a human cell culture system, exhibited an increased tendency to form aggregates, and an increased tendency of alpha-synuclein to co-localize in these aggregates was observed; in elegans, PDR-1 interacts with UBC-2, UBC-18 and UBC-15, homologs of the human E2 enzymes UbcH4/5, UbcH7/8 and Ubc6; PDR-1 also interacts with CHN-1, the homolog of human E4 enzyme CHIP, that interacts with human Parkin and enhances its E3 ligase activity; this C. elegans model recapitulates Parkin insolubility and aggregation similar to several AR-JP linked Parkin mutations.Homo sapiensPaper_evidenceWBPaper00026844
