parp-1 (formerly known as pme-1) encodes a poly(ADP-ribose) polymerase (PARP) orthologous to human PARP1 (OMIM:173870); PARP-1 has two N-terminal zinc fingers that probably bind DNA, and a C-terminal catalytic domain; PARP-1 exhibits poly(ADP-ribose) activity in vitro, which is blocked by mammalian PARP inhibitors; since these inhibitors also cause embryonic lethality in worms exposed to ionizing radiation, PARP-1 is likely to be required in vivo for repairing broken DNA.
