Construction_summaryThis integrated array is composed of plasmids pRF4 [rol-6 (su1006)] (Mello et al., 1991) and pBGSLE (a lin-12::LacZ chimeric gene) (Wilkinson et al., 1994). pBGSLE contains all sequences required to rescue a lin-12(0)mutant (Fitzgerald et al., 1993). A modified lacZ gene (Fire et al., 1990) was inserted into the BamHI site at position 6 in frame to the lin-l2 ATG. The LacZ gene encodes a beta-galactosidase protein that contains a nuclear localization signal and includes its own stop codon but no polyadenylation signal. The mRNA encoded by the lin-12::LacZ gene is therefore predicted to be unstable due to the presence of a large 3' untranslated region containing most of the lin-12 coding sequence. For this reason beta-galactosidase activity is only detectable in the presence of a smg-1 mutation, which stabilizes mRNAs with long 3' untranslated regions (Pulak and Anderson, 1993).
