Interaction_summary | In C. elegans the serotonergic neurons all express tph-1. Comparison of the expression of a tph-1 promoter::GFP transgene in fed WT, unc-2, unc-2; daf-4 and unc-2;unc-43(gf) adult animals grown at 25 centigrades showed WT levels of expression in unc-2;daf-4 and unc-2;unc-43(gf), and relatively decreased expression in the ADF neurons of unc-2, suggesting that unc-2 differences in 5-HT staining were due to transcriptional regulation of TPH-1 levels but not failure to differentiate. | ||||

Remark | To confirm that these changes in tph-1::GFP expression level were not due to a failure of monoaminergic differentiation, the expression of the vesicular monoamine transporter gene cat-1 was compared in the ADF neurons of WT and unc-2 animals using a cat-1 promoter::GFP transgene. Comparison of these strains revealed that their cat-1::GFP expression patterns were indistinguishable, suggesting that the differences in tph-1::GFP expression in unc-2 are caused by the absence of an UNC-2-dependent signal required after serotonergic differentiation. |
