gen-1 encodes the C. elegans homolog of the human GEN1 Holliday Junction resolving enzyme; in C. elegans, GEN-1 activity is required for DNA double-strand break repair and ionizing radiation-induced cell cycle arrest and apoptosis, independent of the CEP-1/p53 pathway, as part of a DNA damage signaling response; GEN-1 exhibits in vitro nuclease activity consistent with it functioning as a junction-resolving enzyme.
