TreatmentSynchronized L1 larvae were grown for three days on plates seeded with RNAi bacteria until control worms (grown on empty-vector control plates) reached the L4 larval stage. L4 worms were washed off the RNAi plates with M9 buffer and Beta-Ala-Lys-AMCA was added to samples to a final concentration of 0.25 mM. Worms were incubated in this solution for 3 hours at 20 degrees Celsius. Liquid OP50 culture was added to the samples and incubated with the worms for 1 hour to wash the Beta-Ala-Lys-AMCA out of the intestinal lumen. Worms were washed 4-6 times in M9 buffer and assayed for intestinal uptake of Beta-Ala-Lys-AMCA by fluorescence microscopy.
