| Construction_summary | Constructs were generated using multisite Gateway cloning (Invitrogen) or with Cold Fusion cloning (System Biosciences). The LOVpep (T406/7A, I532A) and ePDZb1 sequences were codon optimized for C. elegans (except the first 39 bps of ePDZ, because of size restrictions when ordering synthetic DNA). Gateway 5' entry clones: We used a plasmid containing 1.6 kb promoter sequence of wrt-2 (gift of Rik Korswagen). Middle entry clone: The amino-terminal fragment of C. elegans prx-3(aa1-42) was cloned from cDNA into the middle entry clone, based on homology to the fragments used in previous cell culture experiments. | ||
