| Construction_summary | This reporter contains 1.2 kilobases of genomic sequence upstream of the start codon of R09B5.3 and five base pairs downstream. It was obtain by PCR amplification with primers JEP316 and JEP318, containing restrictions site for SphI and BamHI at the 5 and 3 end respectively, followed by cloning into pPD95.75. The pcnc-2::GFP construct (20 g/ml) mixed with transformation marker rol-6 (pRF4) (80 g/ml) was injected into N2. Transgenic animals were identified by the dominant roller phenotype (Zugasti and Ewbank, 2009). | ||
