Construction_summaryThere are three cog-1::gfp fusions made. These fusions insert the gfp coding region into three different positions of pBP150: the 5' end of exon 1; a unique SmaI site in exon 1; and the 3' end of cog-1. The construct with the insertion in the SmaI site was able to rescue the mutant phenotype of sy275. The construct with the 5' insertion did not rescue either mutant allele. The reason for this is not clear. The two exon 1 constructs should label only the cog-1A transcript; however, no obvious difference in the expression pattern was observed between these and the 3' construct. The following describes the expression pattern of an integrated transgenic array syIs63, which contains the 5' insert, unless noted otherwise. To make the 5' construct, PCR amplified gfp coding region was cloned into the AatII site of pBP150, placing the GFP 17 amino acids downstream of the first ATG of cog-1.
