Construction_summaryClone = pKL78. To generate DAF-16a::GFP, the PvuI-StuI fragment from cosmid R13H8, which contains 24.3 kb of the daf-16 genomic sequence, including 6.1 kb upstream of the 5' untranslated region (UTR) and the poly-A signal, was subcloned into plasmid pUC18. A NotI site was created just 5' to the first ATG by PCR amplification of the EcoNI-ApaI fragment, using an inverse PCR strategy. A NotI fragment from plasmid pPD103.87 containing the GFP sequence was inserted into this site to generate an in-frame fusion of GFP immediately upstream of the N-terminus of DAF-16a. One DAF-16a::GFP line (muEx50) was created by injecting DAF-16a::GFP construct at 2 ng/l with 85 ng/l pRF4. muEx50 was subsequently integrated into the chromosome by Gamma-irradiation to generate muIs61.
