Treatment25-50 L1 nematodes were added per RNAi-treatment well (in 24-well plates, 0.25 millilter volume) and the animals were cultured for 2 days at 20 degrees Celsius. Then compound "F17" was added to every other column of clone wells, and DMSO was added to the adjacent columns. The nematodes were cultured for 1 day at 20 degrees Celsius. After the third day of culture, the animals were washed free of bacteria, anaesthetized with sodium azide and imaged. The Nile Red fluorescence of gravid adults was then quantified by integrating the fluorescence intensity of the two anterior-most pairs of intestinal cells.
