Treatment | Worms were harvested by rinsing with S buffer (100mM NaCl, 50mM potassium phosphate, pH 6.0) and resuspended in alkaline hypochlorite (500mM NaOH and 1.2% hypochlorite) until the adults dissolved (510 min). Eggs were then washed five times with S buffer. The eggs were transferred to NGM plates or RNAi-plates, allowed to develop to adults (about 3 days) and moved to fresh NGM plates or RNAi plates (810 worms/plate) supplemented with 40M 5-fluoro-20-deoxyuridine to suppress the production of progeny. The day when young adults were moved was taken as day 0, and the living populations were counted every 2 days. RNAi worms were moved to new RNAi plates every 6 days. Whether a nematode was alive or dead was judged by its response to tapping it on the plate. The experiments were performed at 20 degrees Celsius. | ||||
