[Sundaram M] e1948 recessive suppressor of lin-12 hypomorph, and partial suppressor of glp-1 hypomorph; no effect on lin-12 amorph; no phenotype alone. NA5 (ar23, ar29, ar73, ar77)
[C.elegansII] e1948 : pka sup-25, recessive suppressor of lin-12h hypomorph(n676n930ts), and partial suppressor of glp-1 hypomorph; no effect on lin-12 amorph; no phenotype alone. No suppression of lin-3, egl-15, etc.OA4: ar23, ar29, ar73, ar77. Cloned: encodes predicted 650 aa protein with signal sequence. [Sundaram and Greenwald 1993; GS]
sel-1 encodes two isoforms of a highly conserved transmembrane protein orthologous to human SEL1 (OMIM:602329) and Saccharomyces cerevisiae HRD3, a member of the HMG-CoA Reductase Degradation (HRD) complex that degrades malfolded endoplasmic reticulum (ER)-resident proteins; SEL-1 functions as a negative regulator of the LIN-12/GLP-1 Notch-like signaling pathway in C. elegans where it appears to function cell autonomously to regulate LIN-12 turnover; along with ABU-1, an ER-associated Type I transmembrane protein, SEL-1 may be a component of a cell survival pathway induced when unfolded proteins accumulate in the ER; SEL-1 is expressed throughout larval and adult stages of development, and detected in all tissues except the pharynx; within cells, SEL-1 appears to localize to intracellular vesicles, consistent with its proposed role in protein turnover.
Predicted to contribute to ubiquitin-protein transferase activity. Involved in negative regulation of Notch signaling pathway. Located in cytoplasmic vesicle. Expressed in several structures, including P3.p hermaphrodite; P4.p hermaphrodite; P5.p hermaphrodite; P7.p hermaphrodite; and P8.p hermaphrodite. Human ortholog(s) of this gene implicated in pancreatic cancer. Is an ortholog of human SEL1L (SEL1L adaptor subunit of SYVN1 ubiquitin ligase).
