Disease_relevance | pmk-1 is orthologous to the human Mitogen-activated protein kinases, MAPK14 (p38 MAP kinase) and MAPK11 (p38-beta); studies in C. elegans indicate that the elegans genes pmk-1 and sek-1 (orthologous to human MAP2K3 and MAP2K6) are required for the normal protective role of LRRK2 (leucine-rich repeat kinase 2), during conditions of cellular stress; mutations in human LRRK2 are implicated in late-onset Parkinson''s disease (PD); in an elegans model for human LRRK2 function in vivo, wild-type human LRRK2 complements the function of it''s elegans ortholog, lrk-1, strongly protecting against cellular stress phenotypes in a rotenone (a mitochondrial complex I inhibitor) protection assay; knockdown of sek-1 and pmk-1 in this model, completely abolished the ability of LRRK2 to protect animals against rotenone, this and similar studies with lrk-1, indicate that lrk-1/LRRK2 requires sek-1 and pmk-1 for normal function, linking these MAP kinases to pathologies like PD. | Homo sapiens | Paper_evidence | WBPaper00035592 | |||||
