For eva-1p::EVA-1::GFP, authors first eliminated the stop codon of eva-1, by making the BamHI-SalI fragment by PCR using the following primers: 5'-CCTCTATCATTAAATCATACAATAC-3' and 5'-GGTCGACGGAAAATCATAGTG ACTATAATGTGG-3'. The DNA template for PCR was eva-1 (PstI-SalI), which was made by inserting the PstI-SalI fragment from the cosmid F14B11 into pBluscript-KS. A GFP-encoding DNA was obtained by PCR from pPD95.75, and inserted just 3' to the eva-1 sequence, in which the stop codon and 3' end sequences were eliminated.
