Three genomic fragments carrying different lengths of upstream and similar internal regions of the tmy-1 were fused in-frame with the E. coli lacZ and gfp expression vectors pPD95.03 and pPD95.81, respectively. pTMZIV4349 and pTMGIV4349 carrying 3245 bp PstI fragments of intron three upstream the initial codon were ligated to 1104 bp PstI/BamHI fragment of exons 3b, 4a and 5c. These exons had already been ligated to the BamHI site of pPD95.03 and pPD95.81, respectively. pTMZIV1957 and pTMGIV1957 include a 853 bp BamHI upstream sequence also ligated to the 1104 bp fragment. pTMZIV1219 and pTMGIV1219 are further XbaI deletion constructs to 115 bp upstream from the initial codon. --precise ends. | ||
