| Construction_summary | A genomic fragment of cosmid C50H2 encompassing the entire fshr-1-coding region as well as 3987 bp of upstream and 385 bp of downstream regulatory sequences was cloned into pBluescript KS(+). An NheI site was introduced 51 bp upstream of the fshr-1 stop codon and GFP(S65C) was inserted into this site to generate plasmid pDF97. The plasmid was sequenced for verification. pDF97 was injected into fshr-1(ok778) mutant animals at a concentration of 100 ng/ml to generate fdEx41 extrachromosomal expression line WY335 ( Cho et al,. 2007). | ||
