Subcellular_localizationEGG-1:GFP (pie-1 promoter, N-terminal GFP fusion) appear to localize to the plasma membrane of developing oocytes. The GFP fusion proteins display a swiss-cheese appearance when viewed at high magnification using 3D-deconvolution microscopy. To test for cell surface localization of EGG-1, a live-cell staining assay was developed in which anti-GFP sera was injected into the uterus of hermaphrodites. Anti-GFP staining was observed on the surface of oocytes in the proximal gonad arms and uterus of animals expressing the EGG-1:GFP fusion (C-terminal fusion, GFP on surface). Further, this staining colocalized with GFP fluorescence. No oocyte surface staining was seen with the anti-GFP sera injected into the uterus of hermaphrodites expressing the GFP:EGG-1 (N-terminal fusion, GFP in the cytoplasm). These experiments demonstrate that EGG-1:GFP is on the egg plasma membrane.
