Subcellular_localizationSubcellular localization of vha-8 by immuno-gold electron microscopy using anti-VHA-8 antibodies detected signals in the cytoplasm of the excretory canals and stacked sheets of the plasma membranes of the syncytial hypodermal cells.
TypeAntibodyRabbit polyclonal antibody against recombinant protein. The full-length vha-8 cDNA subcloned in pGEX-4T1 was over-expressed as glutathione S-transferase (GST) fusion protein in BL21 (DE3) E. coli strain, and the fusion protein was used to raise polyclonal antibodies in rabbit.
PatternAntibody staining patterns were consistent with the vha-8::GFP reporter gene expression pattern through all developmental stages, beginning at comma stage embryo to adult. Furthermore, vha-8 expression in canals of the H-shaped excretory cell, was prominently observed in a magnified image. Presumably, VHA-8 is expressed in syncytial hypodermal cells as well as in the excretory cell.
