Subcellular_localizationBy antibody staining: VAB-8S is punctate and roughly colocalizes with thin filaments in preparations double-stained with phalloidin to detect actin. Punctate muscle staining is reminiscent of dense bodies, structures that attach actin thinfilaments to the extracellular matrix. Although not in dense bodies, VAB-8S is near these structures.
VAB-8 was expressed in body wall muscle. In expressing cells, GFP was distributed intracellularly, being excluded from the nucleus.
TypeAntibodyRabbit and mice polyclonal antibody against VAB-8. To raise anti-VAB-8 antisera, amino acids 8581066 of VAB-8 were fused to the 6xHis tag, and purified the fusion protein on Ni agarose from extracts of E. coli. The recombinant protein was injected into mice and rabbits. The resulting antisera was affinity purified against fusion protein coupled to agarose beads, or by binding to gel purified fusion protein immobilized on nitrocellulose strips.

PatternOnly antibodies raised to a region shared by all VAB-8 isoforms detected endogenous protein in whole mounts. In both embryos and larvae, endogenous VAB-8 were detected in each body wall muscle cell but not in any other cells. Muscle cells express VAB-8S. VAB-8L is absent in muscle cells as muscle staining is not observed in animals containing a VAB-8L tagged transgene. VAB-8S localization in muscle cells is punctate. Each muscle cell localizes VAB-8S to its anterior end starting late in embryogenesis.
VAB-8L was expressed in AVKR, and VAB-8S was expressed in the CANs. Other cells expressing VAB-8 included motorneurons in the ventral nerve cord that extend posteriorly directed processes(DBs and DDs), interneurons that extend posteriorly directed axons in the dorsal lateral and ventral lateral nerve bundles (SI and SM classes), and additional unidentified neurons in the pharynx and head. VAB-8 was also expressed in body wall muscle.
