TreatmentApproximately 15 synchronized larvae were grown per well of a flat-bottom 96-well tissue-culture plate in M9 with cholesterol, ampicillin, tetracycline, IPTG, and fungizone. After addition of 80 microliters of induced bacterial suspension per well, the RNAi cultures were grown at 20 degrees Celsius while shaken at 150-200 rpm. After 36-39 hours, the L4 animals were irradiated at 140 Gy with an Elekta linear-accelerator-based system (10 MeV) with an effective dose rate of 6 Gy/min. Four days after irradiation, the number of offspring was scored. Wells containing fewer than ten F1 animals were marked as candidates. Knockdown of essential genes can cause radiation-independent sterility; therefore, a nonirradiated set of cultures was scored as a control. Clones that display radiation-dependent brood-size reduction were repeated in three independent experiments. The identity of all positive RNAi clones was verified by sequencing.
