Interaction_summaryWhen ZYG-9 was depleted from embryos, a correlated decrease in TAC-1 at the centrosome was consistently observed. In embryos having less than 5% ZYG-9 remaining at centrosomes, less than 0.5% TAC-1 (n=3) was observed . For embryos having more than 5% ZYG-9 (average of 7.6%), TAC-1 was detected at the same or lower levels (average of 7.1%; n=10 centrosomes).

Full-length TAC-1 cDNA was obtained by RT-PCR amplification by using primers y54e2a.3BamF1 (5 -CAGGATCCTCGCTCAACACAACCTTCACC-3 (forward) and y54e2a.3XhoR1 (5 -GACTCGAGTTATGCATCCGTCGAAATAACG-3 (reverse) inserted into the pGEX-6-1p vector (Amersham-Pharmacia). Bacterially expressed protein was purified under native conditions by passage through a GST (glutathione-S-transferase) column and was used to immunize rabbits.
