Subcellular_localizationFluorescent signal specific for PME-5 also reveals its presence in the pachytene and diplotene zones where chromatin further condenses into chromosomes. Oocytes at the diakinesis stage, where condensation of chromosome is at its highest, showed the presence of PME-5 on chromosomes. Association of PME-5 with chromosomes is not restricted to the female germ cells. Indeed, nuclei of sperms resident in the spermatheca were labeled with anti-PME-5 antibody. The association of PME-5 with chromatin was observed in a special structure called anaphase bridges. Anaphase bridges are formed during the separation of the two chromosomes. This structure occurs predominantly when dicentric chromosomes are present resulting from the fusion of two telomere-free fragments. This observation further strengthened the possible interaction between PME-5 and chromatin.
One of the most striking features of immunolocalization of PME-5 is its co-localization with chromosomes. This is seen during the early embryogenesis where cell divisions occur for the establishment of the different axis. During the second round division in the early embryos, just prior the cell division of the two founder blastomeres (AB and P1), the condensing chromosomes are clearly labeled with anti-PME-5 antibody.
The distal part of the gonad comprises the mitotic zone, located between the distal tip cell and the transition zone. The germ cells in the transition zone enter the leptotene and zygotene phases, which are the early phases of meiotic prophase, and the DNA in the nucleus starts to condense. The nuclei of these germ cells were visible when probed for the presence of PME-5, suggesting an association with chromatin rather than condensed chromosomes at this moment in oogenesis.
