| Construction_summary | Plasmid pSB120 expressing SNB-1::GFP was constructed from pSB103 (Nonet et al., 1998) by site-directed mutagenesis using the oligonucleotide TTTATgggCTggAggggTACCAgTACACgACCTTT which converted the last codon and termination codon of the snb-1 open reading frame into a Kpn I site. The promoter and coding region was moved as a 3.2 kb HindIII-KpnI fragment into plasmid TU#61 (Chalfie et al., 1994) to yield pSB120. The resulting construct replaces the terminal lysine of SNB-1 with the sequence VPVEK followed by GFP sequences. | ||
