RemarkMutants produce about half the normal number of spermatids compared to controls. Even fewer were produced when tested over deficiency, mDf7. The spermatids that are made appear normal in shape, although they vary in size more than wild type.Paper_evidenceWBPaper00002181

RemarkSpermatocytes fail to complete meiosis; they remain as large, round cells with a granular appearance under differential interference contrast microscopy, and they fail to form spermatids. All defective spermatocytes showed multiple DAPI-positive regions. Nearly half had four condensed DAPI-positive regions, the remaining spermatocytes had more than four DAPI staining regions. Staining intensity was variable. Many spermatocytes showed normal-looking spindles.Paper_evidenceWBPaper00002181

RemarkIn spe-26 mutant spermatocytes, the fibrous body-membranous organelle (FB-MO) complex disassembles in the spermatocytes themselves and the major sperm protein (MSP) filaments disperse. The MO membranes do fold into a compact structure, but they become distended. Spermatocytes show multiple nuclei by electron microscopy and mislocalization of the endoplasmic reticulum (ER) and of ribosomes.Paper_evidenceWBPaper00002181

RemarkMutant hermaphrodites lay unfertilized oocytes. Oocytes can be fertilized by wild-type male sperm. Mutants are completely sterile at 25C but produce various numbers of progeny at 20C and 16C.Paper_evidenceWBPaper00002181
