Construction_summaryClone = pHED.1. Construction of the hsp16-lacZ-GFP fusions. The promoterless reporter plasmid, pPD96.04, was provided by A. Fire (Carnegie Institution, Washington, DC, USA). This plasmid includes the SV40 nuclear localization signal, the S65C GFP mutant, 15 introns, and a lacZ gene equipped with a 3' end from the unc-54 gene. The nematode expression vector pPD16.41 contains the entire lacZ gene. The unc-54 3' end in the pPD96.04 plasmid was replaced with the original lacZ 3' end from the pPD16.41 plasmid. The entire NLS-GFP-lacZ fragment was then blunt ligated into an HpaI-cut plasmid pHS16.48/1B, a pUC13 vector containing the entire hsp16-48/-1 gene pair (nucleotides 2,275-4,189). This placed the hybrid transgene in-frame within the second exon of hsp16-1 (at nucleotide 3,565), and this location was confirmed by sequencing. The resultant recombinant plasmid is designated as pHED.1.
