Fifteen micrograms of total RNA, prepared from wild-type hermaphrodite (lanes A-G and I)or him-8(e1489) XX and XO (laneH) worms synchronized at each developmental stage (embryo, first to fourth larval (L1 stages,and adult), was loaded in each lane. Following electrophoresis and capillary transfer, the blot was hybridized to an antisense riboprobe prepared from a fem-2 cDNA clone. The top panel shows the resulting autoradiogram. The bottom panel shows the ethidium bromide-stained gel before Northern transfer to show relative loading. Lanes A, G, and H are embryo RNA. Lane B is L1, C is L2, D is L3, E is L4, and F and I are adult RNA. The blot in panel B was hybridized with a probe of higher specific activity than the blot in panelA to allow more sensitive detection of the embryonic mRNA.
