Lines of transgenic C. elegans carrying the kin-13:lacZ expression plasmid were generated as described under 'Experimental Procedures.' The histochemical stain for p-galactosidase ('Experimental Procedures') produces an insoluble blue product in cells transcribing the PKClB gene. Panel A shows accumulation of the reporter gene product in adult neurons that contribute processes to the nerve ring and the amphid and labial fiber bundles. These neurons are constituents of the anterior (brackets,1), ventral(brackets,I), dorsal,lateral(brackets,1), and retrovesicularganglia (arrow,2). Interneurons(arrow,3 ) also express the reporter gene. Panel B depicts PKClB promoter activity in neurons of the tail ganglia of an adult. Panel C shows kin-13:lacZ expression in an L1 larval nematode. Wild-type nematodes do not stain under these assay conditions. C. elegans were photographed using Nomarski interference optics with a Zeiss Axioscope microscope at a magnification of x 400.
