WBPicture0000009508DescriptionFigure 2. Localization of mig-2::GFP. (A) mig-2::GFP is expressed in many cells. All animals are wild-type except (iv), and all pictures were taken with a confocal microscope except (ii). Anterior is to the left and ventral is down in all panels. (i) Comma-stage (400 min) embryo. Nearly all cells express the fusion protein, which localizes to the plasma membrane (outlined cells). (ii) L2 animal showing QR descendants (AVM and SDQR), ALM, CAN, and HSN. (iii) Confocal image of AVM, SDQR, and ALM showing plasma membrane localization. (iv) AVM and SDQR displaced posteriorly and paired ventrally in a mig-2(rh17) animal. Note that plasma membrane localization does not change though the cells are displaced. ALM is in its normal position. (v) L3 animal showing the developing vulval epithelium and the anchor cell (AC), which exhibits cytoplasmic staining. (vi) Distal tip cell (DTC; also showing cytoplasmic localization) of the developing gonad in an L3 animal. The distal tip cell is leading the dorsal and anterior turn of this posterior gonad arm. (vii) V cells (lateral epidermis) showing apical plasma membrane localization.(B) Confocal localization of mig-2::GFP in migrating cells in wild-type animals. In each panel, anterior is to the left and is the direction of migration. (i) QR as it migrates over V4. Note the leading edge protruding anteriorly (arrowhead). mig-2::GFP is uniformly localized around the cell periphery. (ii-iv) The daughters of QR, QR.a and QR.p, migrate anteriorly and also exhibit a lamella-like leading edge (arrowheads). (v) After the divisions of QR.a and QR.p, QR.aa and QR.pp undergo cell death (round cells) while QR.ap and QR.pa continue to migrate anteriorly and correspondingly show the leading edge morphology (arrowheads). QR.ap has migrated past its anterior sister cell QR.aa. mig-2::GFP continues to be localized around the cell periphery (with some cytoplasmic staining) in migratory cells.(C) Cell types expressing mig-2::GFP in animals of different developmental stages.
