Embryo and whole mount immunostaining were performed on C. elegans embryos and other developmental stages as described under Experimental Procedures. A, the CPL-1 protein is distributed evenly in early embryos, however, once the embryogenesis progresses; the CPL-1 accumulates in cells of posterior and anterior poles of embryo first (B,arrows) and later in a small group of intestinal cells of the late staged embryos (C, arrows). The CPL-1 is also present in the eggshells (D), and along the length of the L3 worm (E) where it is localized in the cuticular region. Note that the fluorescence signal is very intense at the junction of buccal cavity and anterior part of C. elegans pharynx in the head region of L3 (arrowhead). Distribution of CPL-1 is also seen along the length of an isolated pharynx as well as in pharyngeal gland (arrow) of adult worm (F). The localization of CPL-1 was observed under Zeiss fluorescent microscope using fluorescein isothiocyanate filter sets and photographed at 200 magnification.
