| Construction_summary | [mec-1::gfp] translational and transcriptional fusion constructs. The mec-1 promoter constructs were made by using PCR to amplify 2914 bp of mec-1 sequence upstream of the translational start. The portion of this fragment between PflMI and HindIII was replaced with mini-prep DNA, and the remaining sequence 5' to the PflMI site was sequenced, up to and including the fusion junction. The full-length mec-1::gfp construct containing the mec-1 promoter was made by substituting an MscI site for the stop codon of mec-1 by PCR and subcloning into the promoter fusion. Most PCR-generated sequence was replaced with mini-prep DNA; the rest was sequenced and no errors were found. The full-length construct was tested for rescue. --precise ends. | ||
