A 6.6 kb genomic DNA fragment (from -1,625 to +5,008 nt with respect to the trans-splicing site) of Ce-rcq-5 gene was amplified by PCR on E03A3 cosmid using gene-specific primers (5 -end primer, 5 -TGAGTCCGGTCGACGATTAC; 3 -end primer, 5 -CCCGGGAAGCTTTAACAAATCCATTCTG). Amplified DNA contained the 1.6 kb-long 5 -upstream DNA and the transcribed region, to 10 nt-upstream of the stop codon. Amplified 5 -upstream DNA fragment was cloned between SalI and SmaI sites in expression vector pPD95.75 which encodes green fluorescence protein. --precise ends. | ||
