| Antigen | Protein | The immunogen was a recombinant protein corresponding to the putative large cytoplasmic loop between transmembrane segments 4 and 5 of C. elegans PMR1. The protein was expressed in Escherichia coli using the QIAexpress Type IV System (Qiagen, Hilden, Germany). In a first step the cDNA corresponding to the loop region was amplified by PCR with primers PMR1CYTF (5 -CAAGCATGCCGTGAAGAAGATGCCAGCAG-3 ) and PMR1CYTR (5 -CTAGTCGACTTTTCCCTCCTCAATCGCC-3 ) containing, respectively, a SphI and SalI restriction site at their 5 end. The PMR1CYTF primer corresponds to nucleotides 26812 to 26831 of cosmid CECC4 (accession number Z81490) and PMR1CYTR primer to the inverse complement of nucleotides 136 to 154 of cosmid CEZK256 (accession number Z82088). | ||
