PatternIt is highly and ubiquitously expressed in embryonic cells, although expression was not detected prior to the 30-cell stage, and often appears stronger in the anterior of the embryos. Expression is also observed during the first larval (L1) stage in the nuclei of ventral cord neurons, and in intestinal nuclei, which undergo endoreduplication before each molt. Consistent with the timing of intestinal endoreduplication cycles, UNC-85-GFP is expressed in the intestinal nuclei at later larval stages prior to each molt. From the mid-third (L3) through the fourth (L4) larval stages, expression is observed in vulval precursor cells, which divide to generate the vulval primordial cells. GFP expression is not detected in the germline at any stage of development, or in early embryos, most likely due to germline-specific silencing of transgenes from high-copy arrays. GFP expression was also not observed in adult hermaphrodites, which have completed all of their cell lineages.
