Subcellular_localizationSUR-7HGFP was not localized to the plasma membrane, as was the case for a CDF-1HGFP construct, but was enriched at the nuclear periphery and throughout the cytoplasm in a fenestrated pattern. This localization is endoplasmic reticular for two reasons. First, this expression pattern is indistinguishable from that of a C. elegans YFPHTRAM construct (TRAM is an endoplosmic recticulum (ER) transmembrane protein). Secondly, several CDF family members are known to be ER localized.
TypeReporter_geneThe sur-7HGFP construct is functional since the expression of this construct rescues sur-7(ku119);let-60(n1046).
PatternSUR-7HGFP was expressed in most tissues with the notable exception of intestinal cells. Under a UV-light dissecting microscope, GFP was seen to be strongly expressed in the pharynx, some tail cells and the vulva. Under higher magnification, GFP was observed in many if not all neurons, muscles and hypodermal cells.
