PatternDuring embryogenesis in C. elegans, four asymmetric cell divisions partition germline determinants into the germline blastomeres termed the P0-P4 cells. During these early stages of embryogenesis, HERI-1::GFP was expressed in both germline and somatic blastomeres. In the P1 blastomere, we detected a low level of GFP fluorescence in cytoplasmic puncta whose subcellular distribution and morphology were reminiscent of P granules. By larval stage one (L1), HERI-1::GFP::3xFLAG was seen exclusively in the primordial germ cells Z2 and Z3, and was no longer seen in somatic tissues. HERI::GFP was expressed in germ cell nuclei throughout the remainder of larval development and no GFP expression could be seen in the soma during larval development.
