Subcellular_localizationSMG-4:GFP localizes both to nuclear and cytoplasmic compartments. Confocal microscopic resolution, furthermore, reveals bright puncta both around nuclei and extending into the cytoplasm. In the embryo, GFP luorescence is visible both in the nucleus and cytoplasm of many cells, although the nuclear signal is stronger. Intense GFP puncta can also be seen in or around the nucleus. From successive confocal series through individual nuclei, the very bright GFP puncta appear to be perinuclear, as the signal can sometimes be seen radially distributed around the nucleus. Such perinuclear accumulation can also occasionally be seen by standard fluorescence microscopy. However, in the confocal microscope, additional bright GFP puncta extending into the cytoplasm are also frequently evident.
