TreatmentSynchronized wild-type L1 worms were grown in liquid culture with bacteria expressing dsRNA for 2 days. L4 animals were exposed to 0.14 mM camptothecin (Sigma) in 0.5% DMSO in M9 by soaking for 2 hours. Control animals were mock-treated with 0.5% DMSO. After treatment, the animals were transferred to plates with the original RNAi food. Twenty-four hours later four sets of three worms were transferred to a second plate to allow egg laying. The parents were removed after 18 hr. Dead eggs and offspring were counted 24 hr thereafter.
