Reporter_geneCRISPR line COP1510 mlck-1::degron::mKate2 endogenously tagged with GFP strain, was generated by Nemametrix.
Patternthe CRISPR line showed MLCK-1 expression in the pharynx, uterus, spermatheca, and sp-ut valve. MLCK-1 is seen throughout the spermatheca, including the distal neck, bag, and valve cells. By mid L4, faint MLCK-1 expression is evident in the cells of the developing spermatheca. Expression levels peak in adulthood. During L4, MLCK-1 is localized to apical boundaries of the spermathecal cells, which face the lumen of the spermathecal bag. In adults, MLCK-1 is localized both apically and basally in the spermatheca. As expected, we observed thick bundles of actin at the outer, basal surface with a thinner actin network visible at the apical surface. MLCK-1 fluorescence intensity is brightest at the apical boundaries. However, some MLCK-1 is also localized to the basal cell surface, where it partially colocalizes with the contractile actomyosin bundles.
