Subcellular_localizationSNPC-4:GFP was concentrated in prominent foci only within germline nuclei. These germline-specific SNPC-4 foci were present throughout development of both hermaphrodites and males. SNPC-4:GFP foci colocalized with a marker for chromatin (mCherry:H2B) but not with a marker for perinuclear germ granules (PGL-1:RFP), which is consistent with the expectation that SNPC-4 associates with DNA. SNPC-4:GFP foci were most apparent in the distal population of proliferating and pachytene germ cells and decreased substantially in intensity as proximal germ cells entered oogenesis. Typically, two foci were present in mitotic germ nuclei, whereas only a single, larger focus was detected in nuclei that had entered the pachytene stage of meiosis I.SNPC-4 was found at the promoters of many Pol-II- and Pol-III-transcribed snRNA genes, as well as type 3 Pol-III-transcribed noncoding RNA (ncRNA) genes.
