PatternIn C. elegans L1 larvae, VBH-1 is localized mainly to granules associated to the nuclear envelope of germline precursor cells Z2 and Z3. A weak signal was also detected throughout the entire animal at L1 larval stage that we think might be either somatic cells expression or unspecific binding of the antibody.
In C. elegans, Z2 and Z3 germ cells, as well as the somatic gonad precursors Z1 and Z4, start proliferating at mid-L1 stage. During L2 stage, the resultant germ cell population showed VBH-1 in perinuclear granules. By the end of the L2 stage, the nascent hermaphroditic gonad, including the population of germ cells, divides into an anterior and a posterior population that will form each gonad arm. In L3 larvae, both germ cell populations keep proliferating and extend away from the center longitudinally forming two long narrow groups of cells along the anterior-posterior axis of the nematode. VBH-1 specifically localized around the nuclei of these cells. The final larval L4 stage is characterized by a significant increase in the germline cell population. In the L4 stage, spermatogenesis takes place in the hermaphrodite and male gonads. Germline cells remains as the main site where VBH-1 signal is detected in granules with a perinuclear arrangement.
