PatternExpression of PAT-9::GFP was first detected in the early embryo; by the 2-fold stage of development PAT-9::GFP localized exclusively to the body wall muscle nuclei in each body wall muscle quadrant. PAT-9::GFP continued to be expressed in the cell nuclei of larvae and adults, and it became no longer exclusive to body wall muscle.
The PAT-9 immunostaining of N2 animals showed a similar expression pattern to that seen with the pat-9 transgene. Endogenous PAT-9 was expressedfrom very early stages and throughout all developmental stages, and in adults endogenous PAT-9 was predominantly nuclear and appeared excluded from the DAPI- poor nucleoli. In addition to expression in body wall muscle, PAT-9 was observed in the germ cell-like nuclei of the cytoplasmic syncytium of the C. elegans gonad, where most nuclei are arrested in meiosis.
