To generate pAF15, the putative promoter region of K08F4.7 was recovered by PCR using cosmid K08F4 as a template and primers targeted to immediately upstream of the K08F4.7 and K08F4.6 putative initiator ATG codons. Incorporated into the 5' portion of these primers were restriction site sequences for HindIII and PstI . After purification, this PCR fragment was cleaved with HindlII and PstI and the 'sticky end' produc was recovered by gel purification, and ligated into similarly purified HindIII/PstI-linearized pPD95.69 vector fragment.
