  Construction_summary: egl-17 GFP transgenic animals were obtained by injecting a mixture of 50 ug/ml NH#646 plasmid DNA and 50 ug/ml pMH86 plasmid containing the dpy-20(+) gene into dpy-20(e1282ts) animals. To generate an egl-17-rescuing construct with an expanded promoter region (NH#639), a 12-kb SacI fragment was isolated from the egl-17-rescuing cosmid F38G1 and cloned into Bluescript KSII(+). This egl-17-rescuing fragment contains 10.5 kb of sequence upstream of the initiating codon. To generate an egl-17::GFP reporter with this expanded egl-17 promoter (NH#646), a 3.5-kb SalI to AgeI GFP-containing fragment from NH#293 was cloned into the SalI and AgeI sites of NH#639. The NH#646 construct is predicted to code for the identical EGL-17 GFP fusion protein as described for NH#293, which contains only 4.6 kb of sequence upstream of the initiating codon.
