unc-119(+)] was used. Germline was dissected, fixed and treated with an anti-FLAG antibody. Samples were mounted in a DAPI solution and observed. Representative images from the distal tip cell (DTC) and mitotic region, and different meiotic stages including the early and late pachytene, diplotene, and early and late diakinesis (see Fig.1) are shown. FLAG::CDC-48.1 is detected in the DTC and meiotic stages after the late pachytene stage. The amount of FLAG::CDC-48.1 increases as the meiotic stage proceeds. FLAG::CDC-48.1 localizes predominantly in the nucleus. In the top panels, the open and closed triangles represent the distal and proximal ends of the germline, respectively. Bars, 10 um. (B) Extracts from XA7200 worms expressing CDC-48.1 and CDC-48.2 and XA7203 worms expressing FLAG::CDC-48.1 and CDC-48.2 were subjected to western blotting using anti-FLAG and anti-CDC-48 antibodies.
