We have constructed two -
gt10 cDNA libraries from RNA isolated from a mating population of C. luded 70% hermaphrodites and 30% males among the adults. The average insert gize is 1200 bp with 3.0 x 105 independent clones for one library and 1600 bp with 8 x 106 independent clones for the other. Characterization of the libaries is in initial stages: screening 200,000 plaques of the former library with genomic
fem-3 DNA as a probe, we isolated 8 positive plaques. Screening 200,000 plaques of the latter library with the same piece of
fem-3 DNA, we found 2 positive clones. We are also starting to make stage specific cDNA libraries using RNA isolated from eggs, 12 hr, 24 hr, 36 hr, 48 hr, and 60 hr (25 ) animals. The eggs and staged animals include both hermaphrodites and males. Once these libraries are made, they will be available to anyone for use.
