"To further address whether NHR-25 and ELT-3 coordinate control of their common targets but differentially regulate their own targets, we examined the expression of their specific or common targets before and after depletion of its activities by quantitative PCR (Fig. 6S)... Expression of all four common targets is significantly changed compared with that of the no RNAi control after depletion of either NHR-25 or ELT-3, indicating coordinated regulation of epidermis development between the two. Intriguingly, a cell cycle-related gene,
cdc-6, is apparently inhibited by both NHR-25 and ELT-3, whereas the remaining three targets are activated by the two genes. Expression of three of the four NHR-25-specific targets is significantly reduced by depletion of NHR-25 as opposedto that of no RNAi control, whereas only one of the four is significantly lowered by depletion of ELT-3, supporting the specific regulation by NHR-25. However, it appears that NHR-25 also contributes to the control of ELT-3-specific targets, although no significant binding was identified in their proximity. Expression of three of the four ELT-3-specific targets is significantly altered after depletion of ELT-3, and similar changes were observed after depletion of NHR-25 but at a lower significance level (Fig. 6S).
