COL-19 is an adult-specific collagen, and COL-19 reporter fusion protein is expressed strongly at the cuticle. The COL-19::GFP localization pattern was also changed in the suro-1 mutant background. The spaces between COL-19::GFP stripes were irregular and some COL-19::GFP stripes leaned in the suro-1 mutant.
Monomeric red fluorescent protein (tagRFP) tagged KIN-19 (KIN-19::tagRFP) is expressed abundantly in the worm pharynx. KIN-19::tagRFP expressed in the anterior pharynx undergoes spontaneous age-dependent aggregation. KIN-19 aggregates increased ~5-fold when worms are exposed to 400 mM NaCl for 24 h.
RT-PCR analysis of nph-1 showed a marked decrease in expression of both genes in the daf-19(m86) mutant background compared to that seen in the daf-19(+) background. nph-1::CFP expression was greatly diminished in the daf-19(m86) background compared with daf-19(+).
