fasn-1 was labeled with GFP at the amino terminus (
tn1782) or carboxyl-terminus (
av138). Both alleles exhibited similar GFP expression patterns. GFP is seen in the hypodermis, gonad sheath (esp. proximal) and excretory duct; however, intestine and CAN neuron expression are not evident. Neither GFP::POD-2 nor GFP-labelled FASN-1 is expressed in the DTC. No germline expression of any of these GFP-tagged genes was detected, including by anti-GFP staining of dissected gonads. This was most clearly observed by examination of the distal gonad; however, abundant GFP::POD-2 or FASN-1::GFP expression in the proximal sheath cells made it more difficult to evaluate expression in the proximal germline. The earliest detectable GFP-tagged FASN-1 in embryos appears in developing hypodermis.
