RAB-6.1 is broadly expressed in most tissues, with a large degree of overlapping expression with RAB-6.2. RAB-6.1::GFP is highly expressed in body wall muscle, intestine, somatic gonad, distal tip cells, vulva, and neurons. RAB-6.1 is expressed in multiple GLR-1-expressing neurons, including AVB, AVD, RIG, and PVC, but not AVA or RMDV.
rab-28 is exclusively expressed in ciliated neurons. Both GFP translational reporters for RAB-28 (driven by bbs-8 gene or endogenous promoter) localise along the entire cilium.
We observed NOCA-2 accumulations in several spots in the head and tail of the animal and at the cortex of the epidermal seam cells. Indeed, using the NOCA-2 promoter to express mKate2, we observed a broad expression pattern. Importantly, we detected NOCA-2::GFP in the distal PVD dendrite during neuronal development, which showed a similar localization pattern and dynamics as GIP-2 and RAB-11. In order to validate that NOCA-2 localizes to the MTOC vesicles, we expressed mKate2::RAB-11 in the PVD and observed clear colocalization. In the mature neuron, we could not detect obvious NOCA-2 (nor GIP-2) accumulations at the dendrite tip, which agrees with the loss of distal EBP-2 dynamics.
